Where can I get pCAMBIA3200, pCAMBIA3201, pCAMBIA3300 & pCAMBIA3301?

These vectors have been discontinued because they contain the bar or pat gene encoding the PAT protein (phosphinothricin N-acetyltransferase) which confers resistance to the herbicides phosphinothricin (also known as glufosinate) and bialaphos (bi-alanyl-phosphinothricin).   Commercial formulations of these herbicides are sold under the (trademarked) names BASTA, Buster & Liberty.   We are not authorised to distribute vectors containing this gene.  For questions about licenses, contact Bayer Crop Sciences, and let us know their response in our Discussion Forum.

If you choose to use the bar gene for your work you should seek freedom-to-operate advice before proceeding.  Use of the bar gene or products arising from the use of the bar gene made in or exported to countries where a patent in in force on the bar gene could be infringing, and may stop delivery of your work!

If you have freedom to operate through a license or your work will only be used in jurisdictions where no patent claims covering the bar gene or its use are in force, similar vectors can be made by PCR amplifying the coding sequence out of Streptomyces hygroscopicus and cloning it into the XhoI sites of pCAMBIA1201, 1301 or similar sites that flank the plant selection gene.

When constructing the original pCAMBIA3xxx series vectors we used the following oligos: BarFwd 5'-CAA TCC TCG AGT CTA CCA TGA GCC CAG AAC-3' and BarRev 5'-GAA TCC TCG AGT CAA ATC TCG GTG ACG GGC A-3' with XhoI sites at both ends of the bar gene PCR product to allow direct cloning into another pCAMBIA vector, replacing either the hygromycin or kanamycin resistance plant selection genes.
